Janus Kinase Inhibitors in Dermatology

Clinical Practice
The pathogenesis of alopecia areata includes the secretion of IL-15 in follicular epithelial cells. This activates cytotoxic T cells to secrete IFN-gamma leading to further secretion of IL-15. This cyclic action leads to inflammation and subsequent hair loss.
IL-14, IL-13, IL-31 and TSLP are important drivers of AD pathogenesis.
JAK inhibitor adverse effects can include acne vulgaris, nausea, headache and diarrhea.
